ACTIVE ISOMETRIC TETANIC FORCE F_ACT—
PHYSIOLOGICAL & TEST CELL TELEMETRY

Ready to sample with Length Ratio λ = 0.85 ratio.

0.85 ratio
Preparation: Isolated gastrocnemius muscle (L_0 = 30.0 mm)

Adjust Length Ratio λ, then execute biomedical trial.

Biomedical engineering telemetry and physiological states are evaluated rigorously using 64-bit IEEE 754 precision. Residuals reflect ordinary least-squares regression across successive observations. Continuous data structures support high-fidelity biological and physiological modeling.

Biomedical assumptions and boundary condition limits

The linear model applies strictly to the ascending limb (λ ≤ 0.95); the plateau and descending limbs require piece-wise non-linear modeling. Evaluated on isolated amphibian gastrocnemius muscle (optimal resting length L_0 = 30.0 mm, mass 0.50 g) stimulated at 100 Hz tetanic trains in oxygenated Ringer's solution at 22 °C. Linear ascending limb slope k_m = 4.8000 N/ratio across stretch ratios λ from 0.70 to 0.95.

Biomedical apparatus & physiological sensors.

FOLLOW THE METHOD

A reason behind every step.

Record isometric tetanic force in an isolated skeletal muscle preparation across normalized sarcomere lengths to evaluate the ascending active length-tension slope. Standardized parameter settings: 0.7, 0.75, 0.8, 0.85, 0.9, 0.95 ratio.

01

Muscle Dissection and Organ Bath Mounting

Dissect gastrocnemius muscle with intact Achilles tendon, mount proximal bone clamp in bath, and tie Achilles tendon rigidly to force transducer with braided silk suture.

02

Optimal Length L_0 Determination

Deliver single twitch pulses while adjusting micrometer stage until maximum twitch force is identified, defining optimal resting length L_0.

03

Passive Force Baseline Subtraction

At each length step, record baseline resting tension (passive force F_pass) prior to electrical stimulation.

04

Tetanic Electrical Stimulation

Deliver a 100 Hz, 500 ms train of supramaximal square-wave pulses (25 V, 1 ms pulse width) and record total peak isometric force F_tot.

05

Active Force Computation across Ascending Limb

Calculate active force F_act = F_tot - F_pass across normalized length ratios λ = L/L_0 of 0.70, 0.75, 0.80, 0.85, 0.90, and 0.95, allowing 2 minutes rest between tetani.

06

Ascending Slope Linear Regression

Plot active isometric force F_act versus length ratio λ and calculate ascending slope k_m via linear regression.
